This peptide might be the most powerful one of all. It's got over 50 years of published research and your body is already making it. This peptide stimulates collagen synthesis. It accelerates wound healing. It reduces inflammation in the body and it actually affects the expression of over 4,000 of your genes. It's been studied to regenerate the skin, the hair, the lungs, the liver, the bone, and the gut lining. And this peptide declines naturally as you age.
It's called GHK CU glycil l histadil l g l g l g l g l g l g l g l g l g llysine copper complex and the chances are most of you have never heard about it. Peptides have become mainstream in our vocabulary because of GLP1 drugs like ompic and wiggoi and just this month the FDA announced that it's going to review seven previously restricted peptides for potential reclassification. Now Secretary Kennedy has stated he's a fan of peptides and he uses them himself. And everyone's talking about all different types of peptides, but no one's talking about GHKCU. And this is the peptide with arguably the deepest and most established research base of all. And if you're looking to start peptides, it might be the one that you need.

You're getting it for free and you're making clear decisions about your health and the health of your loved ones. Today we're going to break down GHKCU, my favorite peptide of all. What it is, how it works on the cellular level, what the published research actually shows, how it can help your skin, your hair, wound healing, overall anti-inflammatory activity, and what the contraindications are. And by the end of this episode, you'll understand what GHKCU does in the body. You'll know the research to back it up, and you'll have a framework to evaluate if this peptide is good for you.
What is GHKCU? GHKCU is what we call a tripeptide. It's made of three important amino acids, glycine, histadine, and lysine. And it was first isolated from the human blood way back in 1973 by Dr. Lauren Pickard. And he noticed that when the plasma from young donors was added to liver cells from old donors, the older cells started producing proteins at levels more characteristic of younger tissue and realized that the molecule responsible for that effect was GHK. Now, the miracle of GHK is that it has a high affinity for copper ions. And when you look in the body, it's almost always bound to copper. Well, that's really important because copper is essential. It's an essential trace mineral that plays a role in everything in the body, including enzyatic processes for tissue repair, antioxidant defenses, connective tissue synthesis. GHKCU is that little delivery vehicle that is carrying copper to the cells so it can repair and regenerate. And guess what? Your body produces GHKCU naturally.
Now, for those of you who are listening that are over 20 years old, at the age of 20, the average plasma concentration of GHKCU is 200 nanog per milliliter. By the time you are 60 years old, that's going to drop to about 80 nanogs per milliliter. That's a 60% decline. But here's the crazy thing. That decline tracks really closely to what we observe in aging. loss of skin elasticity, slower wound healing, thinning of the hair, reduced collagen production, increased inflammation. Right? We're not just talking about cosmetic issues. These are measurable biological changes that are starting to reflect the declining capacity for your body's tissue repair and regeneration. So, for decades past its discovery, people were trying to figure out how the heck do we replenish GHKCU externally?
Now you heard of all the different types of peptides, the BPC's, the TB500s, the omellins, the tessamelins of the world. GHKU is actually unusual when you compare them to the other peptides in the world. Because GHKCU has a fantastic foundation of published research. This is one of the most evidence-based peptides in the world. A lot of the peptides that are really popular have really interesting, intriguing animal model data. It's encouraging, but we don't have enough human trial data. Not yet. But GHKCU, that's in a different category. There's in vitro studies, animal studies, human clinical trials. GHKCU has gene expression studies, there are wound healing studies, there are studies on skin density, there studies on hair growth. This is one of the most studied peptides when you're looking at dermatology and regenerative medicine.
One of my favorite things about GHKCU is that when you take a pharmaceutical, it's targeted to a single receptor on a cell. And for the most part, it's affecting a single pathway. GHKCU is a little bit different. It modulates entire networks of gene expression. And the major mechanisms for GHKCU are around collagen, elastin, and the work on the extracellular matrix. I'm going to tell you what that means. Now, the first and most well-known effect GHKCU will stimulate collagen, elastin, and glycosaminoglycans. These are the structural proteins that make up the extracellular matrix of your skin and your connective tissue. You know collagen, you heard of collagen peptides. Collagen provides the structure and the firmness to your skin. Elastin provides the elasticity and glycosaminoglycans like hyaluronic acid provides hydration and volume to the skin. When all three are firing at a high level, your skin is looking amazing. We also know that GHKCU is really potent at stimulating these growth factors that are critical for blood vessel formation now being able to bring more nutrient delivery to damage and aging tissue. One human clinical trial on 21 women showed topical GHKCU showed a 28% average increase in collagen density after 3 months of daily application and this was measured by high resolution dermal ultrasound. The top quartile of participants showed a 51% increase in collagen density. That's incredible. And that's from a single peptide. My partner uses GHKCU cream on her face. It's a thing that has made far and away leaps and bounds the most amount of difference in her skin. Suppleness, firmness, hydration, elasticity. It looks incredible.
Now, secondly, and this is where the research gets more recent, more significant. This is what we're really intrigued by with GHKCU is it modulates the expression of 4,000 human genes. And this is important to understand. In 2010, the Broad Institute of MIT and Harvard created the connectivity map. And it's a database that measures the effects of various compounds on gene expression across tens of thousands of human genes. When the researchers analyze GHK using this tool, they found that it affected 31.2% of human genes at a threshold of 50% change of expression. That means GHKCU is doing more than initially thought. We know that it was affecting your collagen. We know that it was helping your skin look better. But now we're seeing it positively affects your gene expression much more than we ever thought. Thousands of genes are being affected, upregulating genes involved in tissue repair, antioxidant defenses, anti-inflammatory responses, and downregulating the genes that are associated with tissue destruction, fibrosis, chronic inflammation. So, it's affecting your skin positively. It's affecting your genes positively, and it has this potent anti-inflammatory effect. And it's documented. It suppresses potent inflammatory markers like tumor necrosis factor alpha interlucan 6 and it modulates NFCappa beta signaling. And this anti-inflammatory activity for GHKCU is not just relevant for your skin. It's relevant for overall systemic aging. Right? Chronic lowgrade inflammation, some people call it inflammaging, is considered really the hallmark of biological aging. and a molecule that reduces inflammatory signaling while stimulating tissue repair is addressing really the two fundamental aspects of what it means to age.
We also see it's incredible for wound healing. One of the first applications that we know for GHKCU is that it attracts immune cells and endothelial cells to injured sites and it's promoting angioenesis, meaning the growth of new blood cells and stimulating new extracellular matrix to rebuild tissue. This is seen in the skin. It's seen in the hair. It's seen in the gastrointestinal tissue. It's seen in the bones. It's seen in the liver. It's pretty incredible because when it comes to the fountain of youth, this might be one of the closest things. Another thing we see is just the overall antioxidant activity. It modulates the activity of my favorite enzyme, super oxide dismutace. It is the enzyme that neutralizes reactive oxygen species that are made from just any biological reaction, but especially when we're exposed to anything in the environment that is really causing an insult to our system. And we know oxidative stress is another hallmark marker of aging. And GHKCU supports your body's own natural defense systems having a direct effect on free radical scavenging, keeping you younger.
This is one of my favorite studies for GHKCU. A 2022 randomized control trial published in dermatologic therapy enrolled 71 participants and subjects applied a 1% GHKCU cream for 12 weeks. The result was a 55.7% reduction in visible wrinkles compared to baseline. That is incredible. In another separate 2024 randomized control trial with 60 women aed 40 to 65 had a 0.1% GHKCU cream twice daily for 12 weeks. The results showed a 31% reduction in wrinkle appearance, a 28% improvement in skin elasticity, and a 15.6% improvement in collagen density. This is echoed by bigger randomized control trials of 400 plus participants seeing wrinkle reduction at a statistically significant level. What does that mean? If you're using GHKCU at the right concentration for 8 to 12 weeks, you will notice the effects. Now remember, the research showed anywhere between 0.1 to 1% GHKCU was showing positive results for the skin. GHKCU creams can be easily ruined in manufacturing. You want to make sure that the company is following all of the right protocols for producing GHKCU cream. they're doing at the right concentration, which I mentioned, and you want to make sure they're third-party testing.
Now, here is one of my favorite things for GHKCU. If you're losing your hair or it's thinning over the past few years, you have to listen to this. There's clinical evidence that GHKCU is incredible for hair growth. A randomized control trial used a topical formulation containing GHKCU peptide and applied it daily as a spray for 6 months in 45 men with androgenic alopecia, which is male pattern baldness. And the treatment group showed an increase of 52 to 72 additional hairs per square centimeter compared to the baseline placebo group. They showed minimal change for concentration. The 50 mg per milliliter concentration actually outperformed the 100 milligs per milliliter concentration. And that's a significant finding because that means the dose response is called bifphasic. It's not always a more concentration the better. It turns out in this situation that the lower concentration of 50 milligs per milliliter was better. There was a separate 2022 randomized control trial with 50 patients with androgenic alipcia and what they did was compare 0.5% GHKCU to minoxidil which we know as roane. The GHKCU group showed a 22% increase in hair count at 16 weeks compared to 8% with the minoxidil group. And the antigen phase which is the active growth phase in the hair cycle was prolonged by 15% in the GHKCU group. How does GHKCU work? It stimulates the follicle directly. It enlarges the follicle size. It strengthens dermal pill. It promotes angioenesis or blood flow around the follicle and it activates signaling pathways for more growth. And the copper ions also inhibit type 5 alpha reductase which is the enzyme that converts testosterone to DHT in the scalp. And DHT is a primary driver of androgenic alopecia or hair loss. So you actually have a peptide that in randomized control trials has been shown to outperform minoxidal or ro gain. That is incredible. Not only is it producing more hair follicles, it's also keeping the active hair follicles alive longer. It's also extending the growth phase longer in comparison to minoxidil or roane. There needs to be larger trials for hair growth, but the data for sure is promising.
So, GHKCU has a really strong safety profile. Based on the pool data across multiple clinical trials across 12 studies with over 892 participants, they reported mild side effects, some redness, some itching, but really nothing systemic overall. With that said, there are some people who for sure shouldn't use it, and I will preface this. Talk to your doctor first. Individuals with a disease called Wilson's disease, which is a genetic condition where the body can't properly metabolize copper, adding more copper obviously is not going to be a good idea. Individuals with copper allergies who have experienced contact dermatitis to copper may be an issue. Talk to your doctor. Pregnant or breastfeeding, there's not enough data. We don't know. Talk to your doctor. Individuals who are on anti-coagulant therapy, remember GHKCU can stimulate the growth of new blood vessels. So in theory it could be contraindicated. Again talk to your doctor. And let me just say this. If you use GHKCU for the skin, there can be a small amount of people who go through what is called the copper uglies because there have been anecdotal reports of people using GHKCU cream where it appears temporarily that the skin has worsened. And it's because the remodeling process that the GHKCU does on the skin can temporarily for some people make it look worse before it looks better. But it's a remodeling response. It's not a toxicity response and it usually resolves within 2 to 4 weeks.
All right. If you're buying GHKCU, here's what you have to look for. In the clinical trials, we saw GHKCU between 0.1 to 1% concentration. If the product that you buy does not disclose the concentration, that is a red flag. Stay away from it. Now, GHKCU is chemically fragile. I mentioned that before when it comes to processing. It degrades in alkaline conditions. It can oxidize in the presence of free metal ions. The product should be in opaque packaging. It should ideally be an airless pump. That's going to protect it from light and air exposure. And the pH needs to be between 5 and 6.5. You have to talk to the manufacturer and they have to know this already. If they don't know it and you're asking questions and they don't have answers for you, do not buy GHKCU from them. Absolutely should have third-party testing, batch specific certificates of analysis verified by HLC and mass spectrometry. If the company cannot verify this, that the peptide is present in the concentration that they are saying, run the other way. Look for products that are using liposomaal delivery, microotion systems, or technologies that are designed to enhance dermal penetration. If you're going to buy this, avoid any products that list the copper peptide or peptide complex without specifying which peptide is at what concentration. Products in clear glass or plastic packaging can expose the compound to light. Companies that can't provide documentation, stay away from products that are making drug claims like we are treating eczema or curing hair loss. Those are regulatory red flags and those are landmines for companies. Stay away from them, too. And before we close out, go to the show notes. It affects over 4,000 genes in a positive way. It has anti-inflammatory signaling, antioxidant defense systems go up. Wound healing goes up. The safety profile is strong. Of course, talk to your doctor. And yes, the limitations are real. We need more clinical trials, more long-term based data past a year. And the awesome thing is the regulatory environment is shifting. Peptides will be reclassified. And GHKCU is going to become more and more popular. And you're getting ahead of the curve. And remember, in a market that is about to be flooded with peptides, third-party testing is important. A company that is transparent is important. A company that is doing things right.
